What it is
Type II collagen is the primary structural protein of articular cartilage — the tissue covering joint surfaces. UC-II® (undenatured Type II collagen) is derived from chicken sternal cartilage and is processed under conditions that preserve the native three-dimensional structure (undenatured). It works through an entirely different mechanism than hydrolyzed collagen peptides: it is not a building block substrate but an immune-modulating agent.
Why form matters
The UC-II® mechanism — oral tolerance — requires that the protein maintain its native, undenatured structure. In the gut-associated lymphoid tissue (GALT, specifically Peyer's patches), exposure to small amounts of undenatured Type II collagen induces a tolerogenic immune response that suppresses the autoimmune cartilage attack characteristic of rheumatoid arthritis and may reduce inflammatory joint degradation in osteoarthritis. This mechanism requires the specific 40mg dose of undenatured collagen — NOT the 10g dose of hydrolyzed collagen peptides. These two products are not interchangeable despite both being called 'collagen.'
Molecular Forms — What the Research Actually Used
The form in the bottle determines how much actually reaches your bloodstream.
Absorption: Minimal systemic absorption — mechanism is gut-based immune interaction
The patented form. 40mg/day is the established dose. Higher doses do not improve efficacy and may actually impair the oral tolerance mechanism. Must be taken on an empty stomach for optimal Peyer's patch exposure.
Absorption: Variable — depends on processing
Many products claim 'Type II collagen' without specifying whether it is UC-II® (undenatured) or hydrolyzed. Hydrolyzed Type II collagen is a substrate (like Type I), not an oral tolerance agent, and requires the 10g+ dose. Without 'UC-II®' specifically, the mechanism and dose requirement are unclear.
Dosing — What the Research Used
Osteoarthritis — joint pain and mobility
Crowley et al. 2009; Lugo et al. 2016
Rheumatoid arthritis — immune tolerance
Trentham et al. 1993 (original oral tolerance study)
Active individuals — cartilage support
Lugo et al. 2016 (healthy active adults)
Note: The 40mg dose is not analogous to the 10g dose for hydrolyzed collagen. Do not try to use 10g+ of UC-II® — the oral tolerance mechanism may be disrupted by very high doses. Take on an empty stomach in the morning for best Peyer's patch exposure. UC-II® is not a replacement for rheumatoid arthritis disease-modifying therapy — it is an adjunct that has shown immune-modulating benefit in small trials.
Frequently Asked Questions About Type II Collagen / UC-II®
- What is Type II Collagen / UC-II®?
- Type II collagen is the primary structural protein of articular cartilage — the tissue covering joint surfaces.
- What does Type II Collagen / UC-II® do?
- UC-II® (40mg undenatured Type II collagen) was superior to glucosamine + chondroitin (1500mg + 1200mg) for knee osteoarthritis pain in a randomized controlled trial (Crowley et al. 2009) — reducing WOMAC pain scores, stiffness, and physical function more than the standard joint supplement combination. A 2016 study in healthy active adults confirmed significant joint comfort and function improvements. The oral tolerance mechanism is biologically plausible and independently validated in rheumatoid arthritis models.
- What is the typical dose of Type II Collagen / UC-II®?
- The 40mg dose is not analogous to the 10g dose for hydrolyzed collagen.
- Does Type II Collagen / UC-II® interact with any medications?
- Type II Collagen / UC-II® has known interactions with: Immunosuppressant drugs (methotrexate, azathioprine, etc.) — UC-II® works through immune modulation — discuss with rheumatologist before using alongside immunosuppressive therapy, as the mechanisms could interact or complement unpredictably.; NSAIDs / COX inhibitors — No direct pharmacokinetic interaction. UC-II® may allow reduction of NSAID dose in some OA patients — a clinician should evaluate jointly..
- Who should be cautious about taking Type II Collagen / UC-II®?
- Exercise caution or consult a healthcare provider if you are: Poultry allergy — UC-II® is derived from chicken sternal cartilage. May be inappropriate for individuals with poultry hypersensitivity.; Active rheumatoid arthritis on immunosuppressive therapy — discuss with rheumatologist before adding UC-II®.; Confusion with Type I hydrolyzed collagen — the primary safety concern is taking a dose more appropriate for one type of collagen product when using the other (e.g., 10g UC-II® or 40mg hydrolyzed Type I). Read your label carefully..
